As contender advises, a handloader is smart to have a variety of manuals published by component manufacturers with labs and tools we don’t have. A benefit of handloading is the stimulation it provides the shooter who pays attention to his or her marksmanship, firearms, and ammunition, all of it verified, or disqualified, by performance on target.
A 300 grain .45 is a big bullet. Particularly jacketed 300 grain bullet, which instantly implies recoil, since it must be driven faster----or, at least to higher pressure----than a cast bullet of the same weight. There are several reasons for this, which the curious are free to research.
I would remove Unique and other medium burn rate powders from consideration for launching 300 grain jacketed from a .45 Colt. It’s proven that HS-6 (a medium powder) works well with cast .45’s in the 240 to 335 grain range, especially for this shooter deep seated. The inertia of a 300 grain bullet with the friction of a jacket conceptually removes medium----and fast powder----from my consideration. I don’t like to contemplate the TTP picture (Time to Pressure, or pressure curve).
I would first consider 296/H110 (both are Winchester 296)) for 300 jacketed, and then IMR/H4227. Any discussion of pressure is better made with tech crew at Hornady and Hodgdon, where the lab rats collect real data. A 30,000psi faster powder load will give less velocity in standard length barrels due to time under the curve as well. Pressure rises and drops slower with the slow powders, providing more time at higher pressures than a faster peak and burn out of the faster powder

Be aware that their are to Hornady 45 cal 300s, standard and mag. They use the same data, but the mag version has a heavier jacket that won't expand at lower velocity. The standard version will not hold together at 454 or higher impact velocity at close range.

Do yourself a favor, get this and cast big hollow points with it: MP 45 Ruger Only mold. I think you will give up jacketed!

Both are .452 fired into wet newspaper @ 12 ft. The 300 gr HP retained 162 gr. and the 315 gr Cup Point retained 283 gr. Fired from a BFR 454 at approx. 1250 fps. That’s it. I’ve tested it to much higher velocities in other caliber rifles and it is a sight to behold on what it does to vermin. There are falling block 45 Colt rifles out there also to entertain yourself with, but it does well in Rugers.
