I am starting to think about a 500L build on my 44 Ruger Bisley Vaquero and have a few questions with respect to bullet regulation. I'd like to have it built as a 4 5/8 octagon barrel with a dovetailed front sight and rear insert from Dustin Linebaugh. I don't hunt and just shoot big bores for the fun and enjoyment so my loads will likely be in the 950-1,100 fps range with maybe a few full power rounds just to see what they can do. I tend to load heavy for caliber bullets in most of my calibers so the 440 to 525gr class is what I have in mind for the 500. For those who have experience with the 500L, how much difference in point of impact could I expect at 25 yards assuming roughly the same velocity? I was thinking the gun could be regulated to shoot as a point of aim sight picture with a 440gr and I can use a 6 o'clock hold for heavier stuff. I do this all the time with 200gr and 255gr bullets in my 45 ACP revolvers but I have no idea what the difference in POI would be for the 500L. Any help is greatly appreciated!

My 500 L is a custom Bowen with adjustible sites and over time I've run dozens of different load combos thru it with great pleasure. I'm no expert but I can tell you any changes in slugs/powders etc will change POI not only in elevation but windage as well. Adusting for elevation with fixed sites is fairly easy, windage not so much.
My suggestion is begin with a load combination close to the velocity you are comfortable with and see what happens. You should count on trying say half a dozen different loads until you are satisfied with one or two. You will find any changes in components will show up on the target as different POI. Once you find a load that looks promising you can then tackel the job of sight adjustment. One trick I've learned when sighting from the bench is to keep the hands and gun out front of the bags and only rest the wrists and forearms on the bags. This way, when you shoot off hand the POI will be close.
Finding "THE LOAD" can be a labor of love but that's the fun of our sport.

I have an exact build of what your speaking..Dustin will leave you plenty of sight to work with...this gun likes 435 cast preformance at 1100 or so 16 grns of hs6...the 4 inch sherrif model likes 440 balista cast from Montana bullet wks at 750/ 850/fps 10 grns of Trail Boss or 525s From Cast Preformance or Bryan Reece at 10.0 grains of Unique at 850/900 fps 20 yards both same poa... These are both pleasant and potient for most 4 or 2 legged spiecies...

I've been thinking long and hard about this build and when I spoke with Hamilton Bowen a while back the first thing he said was to steer clear of the Vaqueros. He's a wise man but those Bisley Vaqueros look really nice.
500fksjr, any chance you have a picture of that build?

I think everyone who does 5 shot conversions will build them but Hamilton Bowen just mentioned the sight regulation can be frustrating.
