This is one topic I've studied and researched to death. Of the two you mentioned, I'd suggest the 45 Colt simply because it is lighter. The 44 Magnum and 45 Colt (NMBH Loads) are a push in effectiveness. However, the weight difference is substantial.
BTW, I finally settled on a Ruger P90DC 45 ACP as my hiking companion because I needed the ability to reload quickly and the single-action didn't offer that. I happen to "hike" in the same places haunted by illegal border invaders and drug smugglers and can't afford the luxury of carrying what I really want to carry - a Ruger NMBH in 45 Colt.

I'd go stainless vacquero- 45 if you reload, 44 if you don't. I don't go looking for water, but it always seems to find me.
