Both are .45 LC. Supposedly, Bill Grover got the idea for the extra cylinder from the Eastwood movie, "Pale Rider," where Clint carries an extra loaded cylinder with him that he resorts to after his other is spent. Grover was going to have a holster set up for the spare cylinder, to carry it loaded alongside the pistol on the gun belt.
If anyone has an N-frame .45ACP full moonclip that they could send me, I'd like to check it's fit with the cylinder. It would be a hoot to be able to shoot ACP's in it.

If the gun is set up for 45 Colt, you can't get proper headspace with .45 ACP in the same cylinder. .45 ACP in a moonclip sits way too far out the back of the cylinder to work in a .45 Colt cylinder.
