Snyd - Thanks for the info, if I can get 1,050 fps with a 395gr bullet I'd be happy with that result. I don't hunt or live anywhere that requires such firepower, just my personal preference.
Whitworth - Thank you for the input. I'm not searching for top end velocity so I intend to keep loads reasonable. Will probably settle on a heavy slug between 1,000-1,100 fps for my needs if it were a 500. I generally work up a few max loads in each caliber just as an exercise, but don't shoot them all the time.

Ya, 1050 is plenty. Note that I was using 454 brass trimmed to 1.365. I never did try 45 Colt brass with a shorter OAL. Your 395er will probably be shorter though. It'll be interesting to see how it works out for you. So, when are you sending your donor gun to Bowen for your 500L!!??

Well, I've got to pay my final tab on a current project before starting another. I'd guess he'll have a donor on his bench later this year.

Alaskan 454 I have a Bowen Super redhawk in 500 L and a D Linebaugh in 500 L You can find brass easy and there are a good selection of cast bullets available...500L in 450s to 525s and mild to wild loading will do anything you want...That said prior to owning a 500L I had 4/45 colts a 475 L a 480 etc. Now 2 configurations of 500L an one 5.5 in A Horvath 45 colt...My 2 cents....I voted 500 L since you have 45 colt covered

You bunch are the best enablers I've met thus far. I think I talked myself into two customs after corresponding a bit with BCA. One in 45 Colt with an Anaconda Barrel and one in 500L with the custom profile. Now I need to figure out how to pay for them...

Speaking of brass. Someone correct me if I'm wrong but a quick internet search shows 500 JRH brass from Buffalo bore for $130 per 100. Starling 500L brass $134 per 250! No need to cut down brass for the 500L. Plus, plenty of suitable cast boolits available. Plus it's pretty cool to think that one can carry essentialy a 50-70 buffalo gun on your belt!

You have a good point there. I cut down 100 cases for my JRH and it's much easier to just buy the stuff! Also, I just listed some stuff for sale to begin raising funds for this project. I'm probably going to clean out my workshop: singleactions.proboards.com/post/299261/thread

I would go with a 500 Linebaugh. It can easily be loaded from mild to wild and is a real thumper.

I voted .45 Colt. It's more practical for me than the .500 Linebaugh, and I just have no use for any of the other listed rounds. The .500 Linebaugh is neat, but there's nothing you can't kill with the .45 Colt fired from a GP45, or a Redhawk, or a Blackhawk. The 4.5" GP45 is a pretty sweet gun, and I'd highly recommend it. If I could keep only one revolver, it would definitely be a .45 Colt. Whether it would the 4.5" Bowen GP45 or a 4.5" Blackhawk Bisley would be a tougher decision.

Follow Up:
Today I shipped a .44 SRH Alaskan to BCA for Anaconda barrel swap and rechambering to .45 Colt. The search for my PPP in DA form has been years in the making and I believe this will be the one. Ultimately I decided on the .45 Colt for a few reasons:
1. Anaconda barrels look awesome and BCA had them available, was originally leaning towards an SP-profile. 2. 45 Colt is incredibly flexible in a SRH. You can load 155 gr at 600 fps up to 400 gr at 1,200 fps. That's plenty of horsepower. 3. I prefer 6 shots in a DA revolver when possible. 4. Plinking loads are more economical in .45 caliber than .475 or 0.5xx.
Very excited to finally start this project!

Maybe there is something I am misunderstanding in your post?

Never mind. I just read on his site that he can do it on modified Alaskan SRH. I look forward to seeing this when completed!

So this must be record timing, I received an invoice and my Anaconda GP will be shipping back home soon. Very excited for the revolver but need to figure out where that money is coming from to pay the bill!
