With the questions you ask, I would for sure work my way up to what you find tolerable levels for you. I read a lot on the big bore revolvers before I took a step towards them. I had shot a lot of heavy .44's for years, along with about any flavor offered in the TC platform. I found the revolver recoil to be completely different to any of my prior platforms of "hand cannon" shooting. I am ashamed to admit I was a late comer into the 45 Colt. At the upper end with heavy bullets I find it uncomfortable if I do not pay close attention to my grip. I did change grips on my .480 BH, wider at the back strap and sanded and sanded to make them feel right for me. I find it more controllable than the 45 Colt for now.
My thoughts on the large calibers, one can always load down, but safety will not allow one to load so far up on smaller cases.
